Probe bank and key locus service

Project leader: Dr John Crolla

The primary aim of the NGRL’s Key Locus Service (KLS) is to utilise the molecular cytogenetics resources of the Wessex Regional Genetics Laboratory by obtaining probes from:

  1. The Human Genome Mapping Project Resources
  2. Research Collaborators

Within the resources of the NGRL we are able to:

  1. Interrogate the www databases to identify locus specific probes.
  2. Order BACs, PACs, cosmids etc from genome resource centres.
  3. Prepare the selected probes for FISH
  4. Validate the FISH probes either on clinical cytogenetic material and/or by detecting known markers contained within the probes using PCR.
  5. Store glycerol stocks as well as genomic and labelled DNA from all probes

All these activities are funded by the NGRL (Wessex). We are also able to provide a diagnostic service utilising the NGRL FISH probes, and such tests will be charged on a pro-rata basis. FISH tests will normally be performed on peripheral blood cell suspensions which have already been processed for cytogenetic analysis and are in 3:1 methanol:acetic acid fixative. Conventional cytogenetic analysis will be reported from the primary referring laboratory.
